Metodundersökning av tre metoder för kvävemätning i en arktisk äng : Jämförelse mellan mätmetoderna extraktion, inkubation och jonbytesmembran (PRS-sond)

The purpose with this report was to compare three commonly occurring methods for measuring plant available nitrogen in soils. The methods extraction, Incubation and Plant root simulator (PRS) probe - an ion exchange membrane (Western Ag Innovations, Inc., Saskatoon, Canada) method were used and comparison between these methods were conducted. A full factorial experiment were set up in northern Finland with the treatments excluding herbivores, warming and fertilization to see how the inorganic nitrogen content in the soil was effected by the treatments. Soil cores were taken and from them a subsample was incubated for 18 days and thereafter analyzed for inorganic N in laboratory. PRS- probes were in the ground for 1 month.

The role of small businesses is fundamentally important for the economic development in countries. Within this context, business Incubation is used as an entrepreneurship development instrument to promote and support new firms in the early stage of their existence. However, research in the field tends to disregard the perspectives of incubatee entrepreneurs. The performed investigation is based on interviews with entrepreneurs from two different business incubators at Swedish universities. This study uses a Situated Learning Theory (SLT) as a theoretical framework, to examine the entrepreneur's experience of the learning environment in a business incubator.

Within agriculture it is important to optimize the use of nitrogen, not only from an economic point of view but also to minimize damage to the environment. This can sometimes be difficult, especially within organic farming. Organic farmers are using increasing amounts of waste products from the food industry to cover their nitrogen needs. These different organic fertilizers release nitrogen at different rates and under specific soil conditions. Because of this, it would be beneficial to have a standardized method to measure the amount and rate of nitrogen mineralization from organic fertilizers. This project contributes to this knowledge by investigating whether relatively simple and repeatable laboratory methods can be used to estimate the nitrogen fertilizer value of organic fertilizers.

The impact of the distribution in the soil volume of two farm manures on the net mineralization and immobilization processes of nitrogen were studied in an Incubation experiment. Cattle slurry and broiler litter were placed in the soil with three procedures for incorporation including placement on top of the soil (TOP), in the middle of the soil (MID) and mixed homogenously with the soil (MIX). During the two weeks of Incubation, few significant differences in net mineralization were seen between TOP, MID and MIX. However, there was a tendency that MIX led to a larger net immobilization of N than placement at one point (TOP and MID). This is also the result in the literature.

Nutrient effects on microorganism communities in nutrient poor soils

Better, more efficient fertilizers with great productivity and economic returns are needed, but it is important today to find fertilizers that are also sustainable. It is also important to carefully monitor their impact on the environment, including potential side-effects. In this context, the microbial communities that carry out numerous essential functions contributing to a functional ecosystem are of particular interest. The present study is a complement to a pot experiment that investigated the potential of different waste products for use as fertilizers and how they affect the microbial community. In the pot experiment, the clearest treatment change in microorganism community function was found in the fully fertilized treatment used as a positive control. Organisationer emellan, En jämförande fallstudie av företagsinkubation

Business Incubation is an important force of economic development but relatively unheard of in organization theory. This is partly due to the fact that in organization theory, meta-organization or the organization of organizations has been neglected or left lagging as much theorization has been focused on organizing individuals. With the growing number of business incubators, and meta-organizations in general, the need for understanding a relatively new type of organization is prevalent.This case study of the business incubator Global Business Labs Botswana (GBL) aims to clarify the organizational differences among meta-organizations and traditional organizations on a case level, as well as explain the underlying factors for these dissimilarities. With respect to the nature of this case study no general conclusions are drawn from the results. The inference, however, is that the organization of GBL and its incubated companies differs from the traditional organization in five elementary aspects: the weak hierarchical structure, the operational autonomy of its members, the strong focus on recruitment processes, the member identity and the lack of formal sanctions.

Transformation of inorganic mercury (Hg) to methyl mercury (MeHg) is a biotic process that is proposed to be conducted primarily by sulphate reducing bacteria (SRB). The formation of MeHg is of great interest because it is the mercury species that biomagnifies to the greatest extent in the aquatic food webs; about 90 % of all Hg in fish is MeHg. Our understanding of factors involved in the production of MeHg is very important for making a reliable risk assessment. The purposes of this study were to investigate the different factors that control the solubility of Hg and the production of MeHg in a contaminated lake sediment. The lake Ala-Lombolo, Kiruna municipality, is well-known to be contaminated by mercury from different sources. Sediment cores were sampled at three different locations (N, SW and SE) in the lake and divided in three different layers; 0-5, 5-10 and 10-15 cm.

This thesis was carried out to give some background information to a report of SLU (Swedish University of Agricultural Sciences) for the Swedish National Rail Administration (Banverket). The requested information was to provide basic data for an environmental impact assessment of the construction of the tunnel through Hallandsås. The aim with the thesis work was to discuss and quantify the leaching of nitrate and the emission of nitrous oxide from the soil at the groundwater level established through the tunnel project. The nitrogen related processes that could be affected were described and discussed. The different areas where a decreased groundwater level can cause changes in the nitrogen processes are the wet and moist areas.

Incubators have grown more popular in recent years. Studies have shown that companies that have been part of an incubator grow faster and have a higher survival rate. The Swedish government realizes that this benefits the economy. Therefore they have started different funding programs in order to support the incubators. The thesis aims to explore the underlying reasons for why companies that have been part of an incubator often experience high growth.

The purpose of this project is to analyze and study the absorption of the substances ivermectin, spinosad and barettin to hydrophobic and hydrophilic surfaces, and to analyze and study if the barnacle larva are effected when they try to settle on the treated surface.Incubation tests with barnacle larva in Petri dishes were performed as well as absorption tests on hydrophobic and hydrophilic surfaces which were tested with ellipsometry. To determine the surface thickness.The barnacles were placed in hydrophobic as well as hydrophilic Petri dishes that had been incubated with ivermectin, spinosad or barettin. After four to six days the larva was counted to analyze the settling. Dose-related selection of Pradofloxacin resistant Escherichia coli

The study evaluated the Mutant Prevention Concentration (MPC) of Pradofloxacin on three Escherichia coli (E.coli) strains, 2 wildtypes and one first-step gyrA resistant mutant. We also measured the value of AUC (Under the Concentration)/MPC that prevents growth of resistant mutants. It is of importance to reach a concentration above MPC that prevent E.coli from developing resistance against the antibiotic.We used an in vitro kinetic model where we added bacteria? and antibiotic. The culture flask was attached to a pump with an adjustable pump-speed.

Quantification of Tripeptidyl-peptidase II : Optimisation and evaluation of 3 assays

Abstract Tripeptidyl-peptidase II (TPPII), is present in most eukaryotic cells. It cuts tripeptides from the N-terminus of peptides and is especially important for degrading peptides longer than 15 amino acids. TPPII also tailors long peptides into suitable substrates for the enzymes which transport and produce the peptides that MHC I present. Increased levels of TPPII have also been found in certain cancer cells, thus it is of interest to determine if TPPII could be used as a tumour marker.The aim of this study was to optimise and evaluate 3 different methods for quantifying TPPII. Western blot, enzyme-linked immunosorbent assay (ELISA) and fluorophore-linked immunosorbent assay (FLISA) protocols were optimised regarding Incubation times and antibody dilutions.

Utveckling och applicering av en aviditets-ELISA för bovint respiratoriskt syncytialt virus :

Bovine Respiratory Syncytial Virus (BRSV) is a common cause of respiratory disease among young cattle. The virus causes severe losses; the herd mortality rate can sometimes be as high as 20 %. In this study the avidity (i.e the antigen binding force) of BRSV specific antibodies was measured to see if there was a difference between antibodies produced during an acute phase of infection and antibodies produced by earlier infected animals. A commercially available ELISA-testkit against BRSV-specific antibodies was used and an Incubation step with 6M urea was added. The effect of the urea is that it breaks the weak bonds between antibodies and antigen while the stronger bonds remain intact. Four different groups of animals were included in this study; seven calves that were naturally infected, three acutely infected calves with known time of infection, five cows that were seropositive during several years and four experimentally infected calves that had been a part of a vaccine trial. The results of this study showed that antibodies produced during the acute phase of an infection had a low avidity and that the avidity increased with time after infection.

Platelets are important for the healing of damaged blood vessels since they have an importantpart to play in the coagulation process. At the same time, the blood must be kept fluid and notcoagulate at the wrong time. Therefore there are factors that effect the aggregation of plateletsin a positive or a negative way.Previous investigations have shown that platelets during stirring conditions produce reactiveoxygen species (ROS) that weaken the inhibiting effect of nitric oxides (NO) on platelets andthat the drug Dexamethasone (Dex) can reduce the ROS-production.The aim of this project was to investigate if glucocorticoids, in this case Dexamethasone,could restore the inhibiting effect of NO on platelets and if there was any decrease in ROS-production.The result of the ROS-measurements showed a great variance and it was difficult to draw anyconclusions from them, but a clear decrease in ROS, as previous reported, was not shown. In the aggregation experiments the inhibiting effect of NO was observed through the drug S-nitroso-N-acetylpenicillamine (SNAP), a NO-donator.From the aggregation experiments, the result seemed to be that SNAP during longerIncubation time lost its inhibiting effect, probably because the cells become desensitized.With superoxid dismutase (SOD), the effect of SNAP increased, both in the experiment withlonger and shorter Incubation times. Dex seemed to reinforce the aggregation in relation toboth SOD and SNAP.
