Immunoassayer är en väldigt vanlig metod för koncentrationsbestämning av biomolekyler. Denna metod nyttjar antikroppars affinitet för vissa analyter. I detta projekt har det undersökts om Gyros AB:s detektionsinstrument kan göras känsligare. Flera förslag har tagits fram genom efterforskning i artiklar och intervjuer med personer insatta inom området.De metoder som projektgruppen valt att presentera för företaget är enzymbaserad Immunoassay med fluorescerande substrat samt multipel inmärkning av antikroppar, vilket vi tror kommer bidra till att försärka den fluorescerande signalen. Immobilisering av antikroppar med hjälp av traptavidin är ytterligare ett framtaget förbättringsförslag som syftar till att bidra med starkare och förlängd immobilisering av antikroppar till den fasta fasen i Immunoassay.

Optimization of immunoassay parameters in multiplex in the high throughput protein detection technique Proximity Extension Assay
The ability to detect protein-based biomarkers, which are linked to different diseases like colorectal cancer, is very important as a diagnostic tool. Usually complex biological samples like blood are studied which will contribute to different technical issues when performing an assay. The aim with the project is to optimize and develop the high throughput protein detection technique Proximity Extension Assay, PEA, into a 96-plex panel, in hopes of discovering an expression profile for colorectal cancer. PEA was developed by Olink Bioscience and allows specific proteins in a sample to be quantitatively transformed into nucleic acid sequences that are subsequently detected and quantified with real-time PCR. Two proximity probes containing oligonucleotide sequences bind pairwise to target protein and when brought in proximity, a DNA polymerase will extend a hybridization arm from one probe over to the second forming a double-stranded DNA sequence that can serve as a template in real-time PCR.
Seroprevalens av antikroppar mot Encephalitozoon cuniculi hos friska tamkaniner :
The aim of this study was to determine the prevalence of antibodies against the intracellular protozoan Encephalitozoon cuniculi in healthy pet rabbits. Encephalitozoonosis is an important differential diagnosis to ascending infection with Pasteurella multocida in the ears since a common symptom is vestibular syndrome. Samples were collected from healthy rabbits that were brought to an animal clinic in Stockholm for some kind of prophylactic procedure. The samples were analysed by an India-ink Immunoreaction test / Carbon Immunoassay. Of 106 rabbits 7 (6.6%) were seropositve.
Evaluation of specificity of a walnut antiserum and detection of English walnut (Juglans regia) in food with ELISA and Real-Time PCR
Nuts of all kinds are common ingredients in food. For nut allergy sufferers the frequent use of nuts cause problems and "hidden" nuts in food products may elicit allergic reaction when such foods are consumed. Methods for detecting and quantifying walnut (and other nuts) with high sensitivity and specificity are therefore very important.The objective of this project was to verify the specificity of a rabbit antiserum against walnut with immunodiffusion and to determine the size of the dominant walnut antigens with Western blotting. In addition, a commercial sandwich ELISA for walnut quantification was validated and compared with a qualitative real-time PCR.The rabbit antiserum proved to be less specific but after absorption with cross-reacting nuts and seeds it showed high specificity. The ELISA kit reacted, except for walnut, with pecan and slightly with other nuts and seeds tested.

There are several viruses in the programincluding IBDV (infectious bursal disease virus), IBV (infectious bronchitis virus) and NDV(Newcastle disease virus). Method: 96 serum samples were collected from different poultryflocks in Sweden and analyzed by ELISA, which are currently used in the health controlprogram as well as by a commercial prototype of a multiplex Immunoassay manufactured byLuminex Corp., which is currently under evaluation at the United States Department ofAgriculture USDA. This 4-plex assay detects antibodies for the three above-mentionedviruses as well as antibodies of avian reovirus. In the context of this study the ELISAs run inroutine diagnostics as well as a REO ELISA were used as the standard for comparison.Result: The antibody concentration in serum from vaccinated chickens was high while theantibody concentration level in serum from not vaccinated chickens was low.

Prostatacancer är den vanligaste orsaken till cancerdöd hos män i Sverige. Cirka 9500 patienter får diagnosen prostatacancer varje år. Prostatacancer diagnostiseras med hjälp av analys av tumörmarkören prostataspecifikt antigen (PSA) i plasma.Syftet med den aktuella studien var att undersöka den preanalytiska stabiliteten av PSA i plasma och att undersöka hur förändrade provtagningsanvisningar och provtagningsrutiner påverkade analysresultat, arbetsförhållanden och patientsäkerhet.Analysmetoden som användes vid studien var electrochemiluminiscence Immunoassay (ECLIA), vilken nyttjar ljus för detektion av antigen-/antikroppskomplex.I en första studie visades att centrifugerade PSA-prover med icke avhälld plasma, förvarade i 6o C, kan analyseras upp till 5 dagar efter provtagning. Detta till skillnad från nuvarande metodbeskrivning som kräver avhälld plasma vid analys 24 timmar efter provtagning. En andra studie visade att PSA-prov, förvarat i 6o C, centrifugerat och analyserat 24 timmar efter provtagning gav oförändrade PSA-värden jämfört med PSA-prov som centrifugerats och analyserats direkt efter provtagning.
Serum Amyloid A as a possible marker of health and disease in non-domesticated mammals : a retrospective pilot study of SAA levels in dolphins, elephants and tapirs at Kolmården Wildlife Park
The veterinary handling of wildlife, both in zoos and in the wild, commonly requires that the patients need to be captured and immobilized for examination or treatment. This emphasises the need for quick, on-site, laboratory equipment, in order to minimize the analysis time and thus the stress for the animal and also in order to increase the possibility of starting relevant treatment early on.
The aim of this pilot study was to determine whether Serum Amyloid A (SAA) could be a measured in blood samples from bottlenose dolphins (Tursiops truncatus), Asian elephants (Elephas maximus) and South American tapirs (Tapirus terrestris). SAA rises to its maximum about 24 hours after the introduction of an inflammatory agent in companion animals. This also correlates with the estimated time it takes for animals at Kolmården Wildlife Park to be examined and having blood samples collected after being observed as sick. The study also evaluated whether the Eurolyser SOLO, using a turbidometric Immunoassay (TIA) developed for human diagnostics, could be used for such analysis.
This pilot study was a retrospective analysis of SAA in convenience sampled frozen serum, collected from dolphins, elephants and tapirs.
