Development of real-time RT-PCR for the detection of human sapovirus in foods

Food-poisoning is a major health problem and an estimated half a million Swedes are food-poisoned annually, with acute gastroenteritis as a consequence. One of the major causes of contaminated foods is related to food- and waterborne viruses. To be able to trace back the source of contaminant, the method of detecting viruses must be specific and sensitive. No standardized method for detecting foods for sapovirus exists today. The aim of the work described in this bachelor thesis is to implement and opti-mize a real-time RT-PCR method for the detection of all genogroups of human sapovirus in foods.

Purpose: The purpose of this paper is to examine if POC manage to keep cohort interplay between their identity and image.Theoretical framework: This paper is led from theories in area about communicating identity and image. The theories that are relevant to this paper to see the interplay between identity and image is communication theory and brand theories based on associations. From these theories we have composed a model that shows the communication led from identity throughout image in company?s with many distribution channels. It shows what associations that build identity and image and also the complication and threat that the interplay is being exposed to.Methodology: We have chosen to do this examination as a case study.

As the information society becomes increasingly flooded with digital images, the need for efficient image retrieval systems increases as well. To handle the vast amounts of data involved, the indexing process needs to be run automatically, using content-based descriptors extracted directly from the digital image, such as colour composition, shape and texture features. These content-based image retrieval systems are often slow and cumbersome, and can appear confusing to an ordinary user who does not understand the underlying mechanisms. One step towards more efficient and user-friendly retrieval systems might be to adjust the weight placed on various descriptors depending on which image category is being searched for. The results of this thesis show that certain categories of digital images would benefit from having extra weight assigned to colour, texture or shape features when searching for images of that category..

Sjuksköterskors kunskap om och attityder till undernärng

ABSTRACTBackground and aims: Nutrition is a part of the medical treatment, and also a part of the nursingcare. In spite of this, malnutrition is a common problem in the health care sector. Studies show thatnurses have too little knowledge about nutrition and malnutrition and that nutrition has low priorityin the daily work. The aim with this study was to find out the nurses self-reported knowledge aboutand their attitudes towards malnutrition, nutrition screening, prevention and treatment of patientswith malnutrition. Methods: A questionnaire was handed out to 65 nurses in two different care units and 36questionnaires were answered. Results: The respondents from the two care units self-reported their knowledge about nutrition andmalnutrition low. Most of the respondents stated to have a big need of education about nutritionscreening and more than half of the respondents stated that they have a need of education forprevention and treatment of malnutrition.

Quantification of Tripeptidyl-peptidase II : Optimisation and evaluation of 3 assays

Abstract Tripeptidyl-peptidase II (TPPII), is present in most eukaryotic cells. It cuts tripeptides from the N-terminus of peptides and is especially important for degrading peptides longer than 15 amino acids. TPPII also tailors long peptides into suitable substrates for the enzymes which transport and produce the peptides that MHC I present. Increased levels of TPPII have also been found in certain cancer cells, thus it is of interest to determine if TPPII could be used as a tumour marker. The aim of this study was to optimise and evaluate 3 different methods for quantifying TPPII. Western blot, enzyme-linked immunosorbent assay (ELISA) and fluorophore-linked immunosorbent assay (FLISA) protocols were optimised regarding incubation times and antibody dilutions.

Improvement of positive strand assay used in detecting positive and negative RNA of hepatitis E virus

Background: Hepatitis E (HEV) is a small, non-enveloped virus that belongs to the viral genus Hepevirus. HEV is a positive sense single-stranded RNA virus and there is insufficient information regarding its replication. This is mainly because the virus has low capacity to grow in normally used cell cultures. Many specific strand assay detection studies have been done in order to understand more about HEV replication. Unfortunately, these assays have the disadvantage of giving false positive results.Aim: The aim of this project was to improve the positive strand assay to increase specificity and eliminate false positivity which is due to high sensitivity of the polymerase chain reaction (PCR).
