Gene expression from a cold-treated Swedish isolate of Haemonchus contortus

Totally 84 differentially expressed mRNA clones from infective L3 larvae of the parasite Haemonchus contortus, a blood sucking Nematode, were analyzed with single strand hybridization assay (SSH). Altogether 79 clones were sequenced, edited, and compared with proteins found via BLAST in GeneBank. The aim was to investigate gene expression and potential protein expression following storage at 5 °C for 32 weeks. mRNA was extracted from fresh and stored L3. The SSH derived products were cloned into E.

Effects of koccidiae- and nematode infections in first-year grazing calves :

Twenty-four calves, which had not grazed previously, were divided into four groups. They were subsequently inoculated orally with either E. alabamensis oocysts, O.ostertagi/C. oncophora larvae, or both. One group remained uninfected and was used as a control.

There are many factors which affect Nematode communities in arable soil with respect to diversity and population size. This study is based on three different parts, soil samples taken in March 2012 and March 2013, soil samples taken monthly during the growth season of 2013 in bare fallow plots and a pot experiment.

Determination of anthelmintic resistance of Haemonchus contortus to three classes of anthelmintics in a Kenyan sheep flock based on results from faecal egg count reduction test

Haemonchus contortus is the most important bloodsucking gastrointestinal Nematode in small ruminants worldwide. It is often controlled by anthelmintics. Even though other methods to control are available, the most efficient way to treat an animal with clinically manifested haemonchosis is to use one of the available broad spectrum anthelmintics: benzimidazoles (BZ), macrocyclic lactones (ML) or imidothiazoles-tetrahydropyromidines or the narrow spectrum drugs salisylanilides or nitrophenols. Anthelmintic resistance (AR) against all groups of anthelmintics in Nematode parasites has been reported from many countries, and even multiple AR has been observed. In this study I have investigated the resistance levels to all major classes of broad spectrum anthelmintics in a naturally infected flock of sheep of Red Maasai sheep (RM), Dorper sheep (D) and their offsprings on a research farm in Kenya.

There are two species of potato cyst Nematode (PCN), the yellow, Globodera rostochiensis, and the white, Globodera pallida. Each of them can appear in different pathotypes. These are separated by testing the Nematode ability to multiply on a number of potato cultivars with different resistance properties. The aim of this project was to examine the virulence of Swedish and foreign PCN populations and to compare two test methods. Biotests are today the only available tests used to study virulence.

Molecular testing of raspberry plants infected with tomato black ring virus

Nematode-transmitted plant viruses (such as nepoviruses) cause great economically losses in commercially important plants such as tomatoes, grapevines and raspberries all over the world. To avoid spread of the viruses the interest in mapping the viruses has increased. One of the important nepoviruses infecting European red raspberries (Rubus idaeus) is Tomato black ring virus (TBRV). TBRV has earlier been confirmed in raspberry samples from Belarus by ELISA and the aim of this bachelor degree project was to confirm these results by amplifying, cloning and sequencing the coat protein (CP) gene of the virus. In total eight viral cDNA samples, from two different raspberry samples, were amplified and sequenced.

Perch (Perca fluviatilis) is a very popular fish in Sweden and it is common in Swedish waters. In some areas along the Swedish coast the perch population has decreased. It is therefore important to get a view at the health status of perch. During April, July and August of 2008, a total of 346 perches were caught in the Gump Bay in the Botnian Sea. The fish were mainly caught by net but also by rod.

The gastro-intestinal Nematode, Ostertagia ostertagi, and the lungworm, Dictyocaulus viviparus, are the most pathogenic Nematodes of cattle and severe infection can lead to death. They have both a direct lifecycle and on the pasture the parasites spread as freeliving worms. The parasites are mainly a problem for the first season grazers. They have not developed immunity against the parasites and are therefor more sensitive to parasite infections.

Interactions between some plant-parasitic nematodes and Rhizoctonia solani in potato fields

The pathogenic fungus Rhizoctonia solani causes major economic losses for potato producers in Sweden. The producers, as well as advisors, have reported possible increases in severity of R. solani when free-living plant parasitic Nematodes are present and active. There are several examples in the literature of interactions between pathogens where one, or both species, support or increase the damage made by the other organism. These complexes are important to investigate and to be able to forecast, since even low incidences of fungi or Nematodes may result in an interaction of significance.

AbstractThe research project Liberation, supported by the European Commission under the Environment and Sustainable Development Programme (Fifth RTD Framework Programme) aimed at developing a decision support system for ecological risk assessment of contaminated sites has proposed a tiered system including the triad method. This method integrates contaminant chemistry and bioavailability analyses with observed ecotoxicological effects in soil and groundwater. In this study the triad method was used to perform a risk characterization of a former surface treatment and metal industry, now used for the student union buildings of the Mälardalen University in Eskilstuna, Sweden. On the site, high concentrations of copper, zinc and nickel have been detected in the soil in a preliminary investigation of contaminant levels with XRF. For performing the risk assessment of the site according to the triad method, contaminant bioavailability, soil metal concentrations and soil toxicity were determined and an ecological assessment was carried out on four selected soil samples (one reference soil and three samples with low (B), medium (C) and high (D) contaminant levels). Contaminant bioavailability was measured through leaching (ISO TC 190/S7) and biouptake tests (DGT).
