The control of the cell cycle with particular emphasis on the G1/S transition

Tumour development occurs to a large extent because of dysregulation of the cell cycle. Of particular importance are defects occurring in the G1 phase. The reason why G1 is critical is because of the influence of several signals (external signals as well as cyclins and cdks) on this stage. The G1 phase can be divided into two parts, G1pm and G1ps. Each part has its ?own? restriction point which needs to be passed in order to progress to S phase.

Hematologiska analysinstrument för mindre djurkliniker : en utvärdering av QBC-V och Scil Vet abc för analys av prover från hund och katt

Two haematological instruments intended for small animal clinic use were evaluated at the Department of Clinical Chemistry, Swedish University of Agricultural Sciences (SLU), Uppsala, Sweden. The QBC-V and Vet abc were compared to a reference instrument, the Cell-Dyn 3500. Fresh blood samples were obtained from canine and feline patients from the University Veterinary Hospital, SLU. The QBC-V was used for 111 canine samples and 78 canine blood samples were analysed with Vet abc. From cats, 23 samples were analysed with both instruments.

Detta är en litteraturstudie av sjukdomen Devil Facial Tumour Disease (DFTD) som drabbar tasmansk djävul. De lever i vilt tillstånd på ön Tasmanien som ligger utanför Australiens södra kust. Tasmanska djävular är köttätande pungdjur i ungefär samma storlek som en rödräv. De är nattaktiva och lever ensamma, förutom vid konkurrens om föda och under parningsperioden då de möter andra tasmanska djävular. Vid dessa möten blir det ofta slagsmål för att göra upp rangordningen. DFTD ger stora tumörer i munhåla, ansikte och halsregion samt eventuella metastaser.

Hormonell påverkan och receptoruttryck i juvertumörer hos hund

Juvertumörer är de vanligaste tumörerna hos tikar och det finns olika typer av både benigna och maligna juvertumörer. De hormonella fluktuationerna som sker under östralcykeln driver utvecklingen av juvervävnaden och behandling med progestiner (syntetiska ekvivalenter till progesteron) för uppskjutande av löp har kopplats samman med förekomst av juvertumörer, vilket har lett till ett intresse för den hormonella påverkan på juvertumörutvecklingen. Hormonerna är i sin tur beroende av uttrycket av receptorer för att kunna ha effekt i vävnaden. Syftet med den här litteraturstudien är att utreda uttrycket av hormoner och receptorer för progesteron, tillväxthormon (growth hormone, GH) och insulinliknande tillväxtfaktor (insulin-like growth factor, IGF) i normal juvervävnad och i benigna och maligna tumörer, såväl som deras påverkan på juvertumörutvecklingen. De intracellulära signalvägarna för progesteronreceptorer (PR), GH-receptorer (GHR) och IGF-I-receptorn (IGF-IR) är kopplade till cellproliferation och genreglering. De komplexa nätverken av signalvägar interagerar med varandra och en obalans i uttryck och reglering av hormoner, receptorer eller molekyler i cellen kan leda till att tumörer uppstår. Vid progestinbehandling har ökade nivåer av GH och IGF-I uppmätts och högst nivåer sågs i maligna tumörer.

Lymphoma is a group of malignant tumour diseases developing in the secondary lymphatic system. These diseases can develop in all organs as lymphocytes are ubiquitously in the body. In connection to mucus membranes we find mucosa-associated lymphoid tissue, MALT, in which lymphoma can spontaneously but slowly develop, mostly at chronic inflammation or at autoimmune diseases. Today these diseases are incureable with the exception of some cases caused by Helicobacter pylori-infection. Antibiotic treatment of these cases can induce remissions.MALT-lymphomas have characteristic histological and molecular properties.

Canine herpes virus 1 (CHV-1) är en vitt utbredd smittsam patogen. Viruset tycks verka enzootiskt i hundpopulationer och ger upphov till en livslång latens hos infekterade djur. CHV-1 kan sedan reaktiveras vid exempelvis stress, som vid dräktighet. Hos vuxna hundar får infektionen ofta ett subkliniskt uttryck, men kan också ge upphov till respiratoriska symtom samt genitala blåsor och ögoninfektioner. Dräktiga tikar kan drabbas av fertilitetsproblem, med mumifikation, resorption, dödfödsel samt abort av foster.

Review of the litterature and an attempt to evaluate intake levels of iodine and selenium in dogs with and without lymphocytic thyroiditis

The prime aim of this thesis was to investigate whether high or low iodine intake during certain periods of life was, or was not a strong risk factor for the development of canine lymphocytic thyroiditis (CLT). And secondly, if a high or low selenium intake in a combination with high or low iodine intake also may have an influence on that risk factor. To investigate this, a dietary questionnaire was sent out to dog owners. The dogs were selected from an ongoing genetic study of CLT. The questions investigated brand and type of feed during different life stages (puppy, junior and adult) and also if there were any use of dietary supplements.

Evaluation of the retinal ON- and OFF-responses in the dog ERG

The aim of the study was to examine the retinal ON- and OFF-pathways of the canine electroretinogram (ERG). The ON- and OFF-pathways are used to distinguish objects from their backgrounds. A dark object is discernible from a lighter background using the OFFpathway and the ON-pathway is used to descry a light object upon a darker background. This study shows that the dog ERG has both ON- and OFF-responses and that these responses can be separated by prolonging the flash duration over 50 ms. The dog OFF-response waveform was more similar to the cat OFF-response than the human OFF-response (Frishman, 2006; Zrenner and Gouras, 1979). In this study six female three-year-old Beagle dogs were included. Both background and flash stimuli were provided by LEDs (light emitting diodes) with a wavelength of 470 nm, a wavelength that is absorbed almost as effectively by the canine M/L-cones as by the S-cones. These stimuli gave ERGs with both ON- and OFF-responses.

Background. Immunohistochemistry is a useful method in the differential diagnosis between pleural mesotheliomas and metastatic adenocarcinomas in the pleura. Cytokeratin 20 and 7 have been used successfully as markers in studies determining primary location of adenocarcinomas from metastases. The current study is a complementary research of archived paraffininbedded material of cases with cancer origin. This study contributes a bigger statistical material that may facilitate the search for unknown primary site of adenocarcinoma by identification of metastatic cells in the pleura.Methods.

This is a literature study in which I investigate if there are scientific studies that form the basis of the Swedish dog world views concerning the spaying of bitches. The positive impacts are said to be reduced problems with pseudopregnancy, reduced risk of pyometra, reduced risk of mammary and uterine tumors, an earlier cease of bone growth, a more active and happier dog and a bitch who is more tolerant of other dogs. The negative consequences are said to be urinary incontinence, increased aggressiveness, lower metabolism and the increased risk of obesity, a more lethargic individual and altered coat.There are studies indicating that spaying reduces behavioral problems associated with heat and pseudopregnancy. Assuming no ovary tissue persists after the procedure, and there is no progesterone production, the risk of pyometra decreases. Early spaying, before the first heat, reduces the risk of mammary tumors.

Determination of serum thymidine kinase 1 (TK-1) activity is used as a tumour marker in both human and veterinary medicine. TK-1 is an intracellular enzyme involved in a salvage pathway of DNA synthesis. The expression of TK-1 is cell cycle dependent and the activity increases markedly after the G1 to S transition in the cell cycle and then declines rapidly in G2. The pronounced proliferative activity in tumour cells result in a higher TK-1 enzyme activity within the cell. The fact that the increased TK-1 activity can be measured in serum makes TK-1 a useful marker in diagnostics of neoplastic diseases and it provides information regarding prognosis and treatment effectiveness. In human medicine a radioactive based test is used to determine the TK-1 serum activity.

Inhibiting the IGF-1 receptor with the cyclolignan Picropodophyllin: an in vitro study of ovulation, implantation and receptivity in a mouse model

Picropodophyllin (PPP) is an analogue of the anti tumour lignan podophyllotoxin with the unique ability to selectively inhibit the receptor of Insulin like growth factor 1(IGF-1). IGF-1 is believed to play an important part in development of the endometrium facing implantation. With PPP treated mice, studies can be made to measure gene expression from tissue of both treated and untreated mice to compare the role of IGF-1 regarding ovulation, implantation and receptivity. The aim of this study was to analyze gene expression of some steroid hormone receptors and cytokines in ovaries from mice treated with PPP. In this study, seven mice were treated with PPP at different times and tissue was collected.
