Troubleshooting the GFP-tagging gene knockout (GGKO) method for the Leptosphaeria maculans effectors AvrLm6 and AvrLm4-7

An attempt was made to GFP-tag the effector proteins of AvrLm6 and AvrLm4-7 using the GFP-tagging gene knockout method (GGKO) developed by Saitoh et al. (2008) in order to determine whether or not they are secreted. Successful pETHG-(target)KO vectors were not generated. The protocol was examined for potential errors. Fatal errors were pinpointed to the ligation reaction and the transformation required to generate and propagate the desired vector pETGH-(target)KO.

Phycoremediation of pesticides using microalgae

Every year, pesticides are found in surface and ground waters in Sweden. Fungicides are in common usage and applied in high amounts against potato late blight. The present thesis examined the possible removal of four fungicides (metalaxyl, cyprodinil, propamocarb and mandipropamid) from water using the microalgae Chlorella vulgaris. Microorganisms are capable of decomposing a range of organic pollutants and the main focus in previously published studies has been on Bacteria and fungi. Microalgae are mostly studied due to their high capacity in biosorbing heavy metals.

Prevalence of selected infectious diseases in Samoan dogs

SCA is the largest private owned forest owner in Sweden, with an ownership of 2 million hectares of productive forestland. To manage the planning of such large areas and to fit the traditional organization structure of a Swedish forest company, the planning approach is subdivided into three stages; strategic, tactical and operational. The strategic part, based on a statistic sample of stands, returns a harvest level estimate which should then be implemented at actual sites in the tactical part. This procedure can lead to incompatible solutions as the strategic solution is not necessarily possible to translate into actual sites in the tactical plan. The geographic aspect is important in tactical planning due to the large costs for moving equipment and maintenance of a large number of forest roads.

The focus of this study are small sewage treatment plants which use a technique where mechanical, chemical and/or biological reduction of pollutants is being used in the same facility to reduce nutrients in household wastewater.The aim of the study is to investigate how different municipalities ensure that small sewage treatment plants fulfill the requirements and reduce pollutants as efficient as the manufacturers claim.

Membranbioreaktorteknik- en framtida lösning på Lundåkraverket? : Membrane bioreactors- A future solution at Lundåkra wastewater treatment plant?

Membrane bioreactors (MBR) combined with biological phosphorous treatment (Bio-P) has been investigated if that could be an alternative to the future expansion with conventional active sludge treatment at Lundåkra wastewater treatment plant (WWTP) in the proximity of Landskrona, Sweden.The results showed that the surface demand in the biological treatment will be reduced with 67% with the MBR-alternative compared to a conventional activated sludge process. The nutrient removal in the MBR-solution was decreased or unchanged for nitrogen-, phosphorous- and organic parameters. However, the energy demand will increase with an estimated cost of approximately 1 million SEK annually. Moreover, the extra chemical consumption is estimated to cost 370 000 SEK annually.The disturbances caused by filamentous Bacteria such as sludge dispersal could be resolved with MBR-technology because the membranes are not affected by these organisms. The Bio-P process combined with MBR-technology works if the wastewater recirculates from the membranes to an aerobic zone.In the proposal, there will be a new pre-treatment step, for instance hole screens.

The purpose of this report is to investigate the possibilities with drain water heat recovery (DWHR) in residential buildings. Information and relevant theory has been collected and summarized in this report. Calculations have been done for given scenarios to evaluate profits. DWHR heat exchangers use simple technology and have long life-time. The heat exchanger is connected to outgoing drainage pipe and incoming cold-water supply so countercurrent flow is accomplished.

Effect of Bacillus mediated priming on different natural genetic variants of Arabidopsis thaliana

In two feeding experiments Bacillus induced priming of plant defense was tested for its effect against the generalist insect pest Spodoptera littoralis. The main hypothesis for the study was that Bacillus induced priming would enable the plant Arabidopsis thaliana to defend itself better against this insect herbivore since this priming seems to involve jasmonic acid, known to be important for plant defense to insects. A secondary objective of the study was to examine if any differences in a primed defense capability could be seen between different natural genetic variants (ecotypes) of A. thaliana reflecting habitat differences in pest pressure. The methods used for plant cultivation and Bacillus inoculation were aimed at reflecting conditions that can be expected in an agricultural cropping system. This meant that all plants were cultivated in soil, and the feeding experiments were conducted with the plants growing intact in the soil system.

Otitis externa is a disease regularly encountered in veterinary practice. It is estimated to be the most common disease in the external ear canal of dogs and treatment is often delayed due to unfortunate pet owner ignorance. Owners tend to assume that scratching of the ears and head shaking are natural behaviours in their dog and may be neglecting the problem unknowing of how severe it actually is. A literature study has been carried out to examine the causes of otitis externa in order to present a collection of measures that can be used in the treatment and prevention of the disease. In turn these measures can be advised to the dog owner and used in the home environment to make the best qualifications for healthy ears in the dog. The causes of otitis externa have been divided into predisposing, primary, secondary and perperuating causes.

The purpose of this study was to investigate whether resistant maltodextrin is suitable as a fiber source in the probiotic fruit drink ProViva Mango without affecting the probiotic bacterium Lactobacillus plantarum 299v, viscosity or the sensory experience of the drink. Resistant maltodextrin is produced from hydrolyzed corn starch and is classified as a soluble dietary fiber. It is relatively resistant to digestion in the small intestine, where approximately 10% can be broken down, 50% can be fermented in the colon by the intestinal flora and 40 % is excreted in the faeces. Resistant maltodextrin has several important qualities when it comes to food processing. When added in a product, it does not affect the viscosity or taste and it is heat and acid stable.
